RESUMO:

Diabetic nephropathy (ND) is complication of Type 1 Diabetes mellitus (T1DM). The loss of podocytes from the renal glomerulus is an early and key event in the development of ND. The technique to concentrate the urinary exosomes eliminated in the urine has allowed the quantification of small concentrations of cellular components carried in these extracellular microvesicles, rich in biomarkers, among them proteins. Among the proteins found in urinary exosomes are: Wilms-1 tumor protein (WT-1), which plays an important role in the development of renal diseases; nephrin and podocin, key proteins in maintaining the integrity of the glomerular filtration cleft.
Objective: Thus, this study aims to evaluate the WT-1, nephrin and podocin proteins involved in slit diaphragm of the glomerular filtration from renal cortex and exosomes urinary, which may represent potential early and non-invasive markers of glomerular damage and of ND.
Materials and Methods: The animals were divided into 2 groups, diabetics induced by Strepzotocin (STZ) and control, being evaluated in 2 periods (30 and 60 days), regarding body weight and biochemical parameters such as glucose, urea, albumin and creatinine.
Results: Reduction in body weight and serum albumin was observed in the diabetic group when compared to the control group in both periods. There was also an increase in serum glucose and urea concentrations in these same groups. In addition, classic symptoms of T1DM, such as polyuria, polyphagia and polydipsia, were observed.
Conclusion: Thus, the T1DM-induced by STZ was confirmed by hyperglycemia and classic symptoms, such as body weight loss, polyuria, polyphagia, and polydipsia. In addition, changes in concentrations of serum urea and albumin suggest a possible initial renal damage in T1DM.
