SUMMARY:

Oral mucositis is a side effect, frequent and debilitating in patients with head and neck cancer who are undergoing treatment with chemotherapy and radiotherapy. Characterized by ulcerative lesions in the oral mucosa, OM causes impairment of the patient's well-being as well as limits anti-tumor treatment. Since there are few protocols that have demonstrated effectiveness in preventing this side effect, this study aims to analyze the effect of gold nanoparticle (NpAu) in oral mucositis induced by 5-Flourouracil (5-FU), in hamsters studying signaling pathway. In this experimental model, the animals underwent intraperitoneal injection on the 1st and 2nd day at the respective doses of 60 and 40 mg / kg. The right oral mucosa received superficial grooves on the 4th day (mechanical trauma). The animals were euthanized on the 10th day of the experiment, the samples were then collected. The experimental groups are composed of: (PVP), (TM) (5-FU) (NpAu 62, 125 and 250µg / kg. IL-1β and TNF-α were measured by ELISA. Glutathione (GSH) Immunohistochemistry analyzes for NF-kB and COX-2 and gene expression for NQO1, HMOX1 and KEAP1 by RT-PCR The results showed that NpAu at a dose of 250µg / kg reduced the levels of IL pro-inflammatory cytokines -1β and TNF-α (p <0.05), and increased GSH levels.The result of immunohistochemistry showed a reduction in immunostaining for NF-kB and COX-2 and increased the gene expression of NQO1 and HOMOX1. Thus, it can be concluded that the gold nanoparticle demonstrated a protective effect on oral mucositis induced by 5-FU in hamsters, by modulating the signaling pathways, with an anti-inflammatory and antioxidant effect.
