RESUMO:

The Zika virus is an arbovirus of the genus Flavivirus, recent epidemics have presented as main consequences the increase in the number of cases of Guillain-Barré syndrome and the emergence of the congenital syndrome of Zika virus that can result in microcephaly and other neurological damages. Immunological mechanisms that confer protection or pathology during infection by this virus have not been elucidated yet. Thus, the objective of the present study was to evaluate the profile of the cellular immune response in patients during the acute infection by Zika virus. The expression of messenger RNA (mRNA) of transcription factors and cytokines related to the different differentiation profiles of CD4 T lymphocytes (Th1 / T-bet-IFN-γ and TNF-γ, Th2 / GATA-3 / IL-9, Th9 / PU.1 / IL-9, Th17 / RORt / STAT3 / IL-17, Th22 / AHR / IL-22 and regulatory T cell / FoxP3 / IL-10 and TGF-β). Peripheral blood mononuclear cells (PBMC) from 30 patients with symptoms of acute Zika virus infection with diagnosis confirmed by real-time RT-PCR (qRT-PCR) and nine uninfected healthy individuals were used to quantify the immunological response profile cell phone. Patients with acute Zika virus infection had high expression of PU.1, RORt, IL-22, FoxP3, IL-10 and IFN-γ when compared to healthy controls. On the other hand, patients infected by the zika virus showed reduction in GATA-3 expression. Similar expression of AHR, iNOS, IL-17, IL-9 and TGF-β was observed between the group of patients infected with zika virus and control subjects. Our results indicate that acute infection (up to 5 days after the onset of symptoms) by the Zika virus induces the production of IFN-γ, IL-22 and IL-10, cytokines produced by Th1, Th22 and regulatory T lymphocytes.
