RESUMO:

Chikungunya virus infection causes high local morbidity, mainly arthralgia and
arthritis. An inflammation induced in the joints by viral infection involves innate and
adaptive immunity. However, the immunological mechanisms involved in protection
or pathogenesis are not yet extended. Thus the aim of the present study was
evaluated as expression of innate immunity receptors and cytokines induced by
their activity in patients infected with Chikungunya virus. In this case, real-time
PCR expression of Toll receptors (TLR3, TLR7, TLR8, TLR9), RLR (MDA5),
Adapter molecules (TRIF and MyD88) and cytokines (IFNα, IFNβ, IFNγ, IL -6, IL-
12 and TNFα) in PBMC from patients in the acute phase of Chikungunya infection
(N = 28), uncontaminated (N = 9) as experienced as negative controls. The
CHIKV-infected patients presented the mRNA expression of TLR3, TLR9, MDA5
and IFNα, in contrast, reduced expression of TLR8 mRNA, when compared to the
healthy control group. In addition, positive correlations were observed between
MDA5, TLR3, TLR7, TLR8 and TLR9 mRNA expressions in relation to IFNα.
Interestingly, positive correlations were also observed between MDA5 and TLR3
mRNA expression with IFNβ. Our results indicate that CHIKV infection in patients
induces elevated production of IFNα, IFNγ and IL-6, antiviral microbicidal
molecules of innate immunity, likely due to the recognition of the virus by MDA5
and TLR3, as well as mechanisms that, TLR9 collaboration.
