RESUMO:

Chikungunya fever (CHIKF) is an arbovirose transmitted by mosquitoes of the genus Aedes, mainly by Ae. aegypti and Ae. albopictus. Its etiologic agent, Chikungunya virus (CHIKV), is an alphavirus with single stranded RNA genotype and positive polarity. This virus remained silent for almost half a century however by the 2000s it gained notoriety because it was the cause of an emerging disease that affected different regions of the world.Knowledge about the role of the immune response, both in control and in the pathogenesis of the disease, are not well established and the available data are scarce. In this study we evaluated the profile of the innate immune response presented by patients with CHIKV acute infection, by quantifying the expression of Toll-like receptors and their adapter molecules; MDA5 receptor; type I interferons (IFNs); cytokines IL-6, IL- 12, IFN-γ and TNF-α, in PBMC, through qRT-PCR. Were analyzed blood samples from 28 patients with previous diagnosis of CHIKF in the acute phase and nine blood samples from healthy individuals, used as a control group. After an analysis of all the innate immune response receptors and adapter molecules, it was possible to observe that only TRL3 and TRIF had significant expression.Relevant, but not significant, levels of expression were observed for TRL7, TRL9 and MDA5 in infected patients, when compared to the negative control group. Regarding the cytokines, only IFN-α presented significant expression.When compared to the negative control, it was found that there were levels relevant for IL-6, IL-12 and IFNγ; and low levels of expression for IFNβ and TNFα, however none of these results were significant. A significant correlation was observed between TLR3 and TRIF. Correlation was also observed between TLR3, TLR7, TLR9 and MDA5 with IFN-α; as well as between MDA5 with IFN-β, IFN-γ, IL-6 and IL-12; as in TLR3 and TLR9 with IL-12. In addition, correlations were found between IL-12 and IFN-γ, and IL-12 and TNF-α. The results suggest that, in patients with acute CHIKV infection, the MDA5 / IPS-1 and TLR3 / TRIF signaling pathways of the innate immune response were more important than MyD88-dependent signaling.
