SUMMARY:

Cashew (Anacardium occidentale) is a native plant from the Northeast region of Brazil commonly used in folk medicine due to its anti-inflammatory, astringent and healing properties. However, it is also used for clandestine induction of abortion, with little information on the effects related to its use during pregnancy. Knowing that the use of plant extracts during pregnancy can cause both maternal and fetal toxicity, the objective of this study was to evaluate the toxic and teratogenic effects of aqueous extract (EAq) of (A. occidentale) bark using Zebra fish as an experimental model. Embryos were exposed to EAq at 6, 12, 24 and 48 hours after fertilization (hpf). Adult females were also exposed to verify the effects of EAq on fertility and behavior. Experimental procedures were previously authorized by CEUA / UFRN, and acute toxicity tests on (D. rerio) and adult female embryos followed OECD recommendations. To determine the median lethal dose (LD50) in the embryos, male and female progenitors were used, in a 2: 3 ratio. After spawning the eggs were collected and stored in Petri dishes for exposure to different concentrations of the extract previously diluted in 10 ml of water. After the exposure period, the eggs were washed and stored in 24-well plates for stereoscopic observation for 24, 48, 72 and 96 h. Thus, the estimated LD50 for the 6, 12, 24 and 48 hpf stages was: 311.8 mg / 10 ml; 308.8 mg / 10 ml; 199.5 mg / 10 ml and 91.0 mg / 10 ml, respectively (p <0.05). For determination of LD50 in adult animals, 15 individuals remained for 96 h in 1 L aquariums containing the diluted extract in different concentrations, and the LD50 of 5.4 mg / L of cashew bark EAq was estimated. During exposure of embryos to exploratory doses to estimate LD50, it was possible to observe effects indicating toxicity, such as chorionic membrane deformation at all stages exposed to doses above 200 mg / 10 ml EAq, and a delay in hatching rate. at 6 hpf stage exposed to 50 mg / 10 ml of EAq. A higher frequency of hatching delay was observed than expected (X2 = 6.93; p (Fisher) = 0.009 and gl = 1), so this effect depends on the exposure to the extract. Tests are being performed with adult females to observe the toxic effects of EAq in two treatments (chronic and acute), and tests with embryos exposed to therapeutic doses to verify the presence of malformations.
