Ten years ago, an outbreak of the Zika virus (ZIKV) generated thousands of patients with different levels of malformations and clinical consequences that still challenge healthcare professionals. One of the characteristics of the syndrome is that the severity of the consequences diminishes as gestation enters the third trimester, and almost none of the postnatal cases develop a central nervous system (CNS) infection. The beginning of the third trimester represents the transition from prosencephalic neurogenesis to gliogenesis. One of the main glial cell types, the astrocytes, contributes to the formation of barriers between the CNS parenchyma and blood or meninges, as well as forms the glymphatic system. The initiation of the glymphatic system's (GS) function marks a shift in CNS fluid dynamics, characterized by the onset of drainage of cerebrospinal and interstitial fluids. Here, we demonstrate that ZIKV brain infection, both before and after the formation of the GS, results in distinct clinical, physiological, and histological outcomes. The mouse brain becomes more resistant to ZIKV infection after the formation of GS. The brain susceptibility window to ZIKV, which closes by the onset of the GS, can be reopened by the pharmacological inhibition of the brain arterial contractility, which is fundamental for liquid flow through the perivascular space. Therefore, the GS is part of the innate immunity of the CNS to ZIKV, and the onset of its function influences the susceptibility of the embryo to the congenital ZIKV syndrome.
