Evaluating the Efficiency of Phytol Nanoemulsions in Modulating Anxiety-Like Behavior in Female Rats

SUMMARY:
Phytol is a diterpene with effects on the central nervous system. The literature shows
anxiolytic effects of this branched and unsaturated chain alcohol in Swiss mice
exposed to the elevated plus maze. However, studies investigating the effects of
nanoemulsions of this compound in animal models of anxiety are lacking. Thus, this
study aims to examine the effects of intraperitoneal administration of phytol,
incorporated into a sunflower oil nanoemulsion, in rats exposed to the elevated plus
maze. The nanoemulsion was prepared using the phase inversion method, varying the
concentration of phytol from 0.1 to 0.5 grams. The stable formulation containing the
highest concentration of phytol was subjected to physicochemical characterization,
which included evaluating droplet size, polydispersity index, zeta potential, and pH
over one month. Fourier Transform Infrared Spectroscopy, Transmission Electron
Microscopy, and Chromatographic Analysis characterized the nanosystem. The
formulation was subjected to hemolytic evaluation evaluatingand in vitro DPPH test. In
vivo effects were evaluated in female Wistar rats using the elevated plus maze test.
The animals were divided into groups according to their treatments (vehicle; Diazepam
2 mg/kg; phytol 25 mg/kg; phytol 75 mg/kg; blank nanoemulsion; phytol nanoemulsion
25 mg/kg, and phytol nanoemulsion 75 mg/kg) administered intraperitoneally.
Characterization data revealed that the highest concentration of phytol for
nanoemulsion formulation was 0.25g, presenting an average droplet size of
approximately 160 nm, a PDI of 0.235, and a negative zeta potential of -27 mV, with
stability for more than 30 days. A significant reduction in anxiety-like behavior was
observed in the animals, with increased percentages of entries and time spent in open
arms, after treatment with both blank and phytol-containing nanoemulsions at both
concentrations, compared to the vehicle and free phytol groups at a concentration of
25 mg/kg. The promising anxiolytic effects of the formulation suggest greater efficiency
of the nanosystem compared to the free drug.
