RESUMO:

Leishmania infections can have different clinical outcomes, varying from symptomatic disease to self-resolved infection  without apparent clinical symptoms. Genetic variability among Leishmania isolates may affect these outcomes. The aim of the  present study was to examine the genetic variability at genome level of multiple Leishmania infantum isolates from human symptomatic and asymptomatic visceral leishmaniasis (VL) cases and from dogs in an endemic area for VL in Northeast Brazil. For this, the genomes of 20 different isolates, distributed across 2 distinct species (dogs and humans) and clinical phenotypes (asymptomatic and symptomatic), were cloned, sequenced and mapped to the L. infantum JPCM5 reference genome. Time and distance were considered as main variables for distinguish isolates’ groups. Single nucleotide polymorphisms (SNPs) in both coding and non-coding regions, and were assessed based on quality values. Phylogenetic trees was calculated based on a Bayesian grouping method as the same way as population dynamic analysis. Ploidy values for each chromosome was calculated based on read depth coverage. Our results showed a high identity pattern among all Rio Grande do Norte isolates for L. infantum, in contrast to other countries isolates’. Despite this result, genomic variation and phylogenetics analysis showed a grouping tendency, according to isolates’ features. In addition, it could also be observed that the rate of diversification in parasites population had increased since the 90’s and it may be associated to the ploidy variation at different chromosomes and isolates.
