RESUMO:

Fucan is a term used to denote a family of sulfated L-fucose-rich polysaccharides. We have extracted sulfated polysaccharides from the brown seaweed Dictyopeteris delicatula by proteolytic digestion, followed by separation into six polysaccharide families by sequential acetone precipitation. The chemical analyses demonstrated that all polysaccharide contain heterofucans composed mainly by fucose, xylose, glucose, galactose, uronic acid, and sulfate. They were evaluated for their anticoagulant, antioxidant and antiproliverative in vitro activities. The fucan F0.5v and F0.7v at 1.0 mg/mL showed high ferric chelating activity (~45%), whereas fucans F1.3v (0.5 mg/mL) showed considerable reducing power, which about 53.2% of the activity of vitamin C. No clotting inhibition was observed in PT test of any of the samples at the concentrations assayed. On the other hand, APTT were prolonged by all the polysaccharides, in a dose dependent manner. The fucan F1.5v presented the most prominent anticoagulant activity; it showed similar anticoagulant potency as enoxiheparin, at same concentration. The best antiproliferative activity was found with fucans F1.3v and F0.7v. The antiproliferative effect of fucan F0.7v was a dose-dependent, reaching the saturation around 0.4 mg/mL. The heterofucans F1.3v also showed a dose-dependent antiproliferative activity, reaching the saturation around 2.0 mg/mL. However, its activity was very higher than F0.7v inhibiting almost 100 % of HeLa cell proliferation. Several fucans demonstrated promising antioxidant, antiproliferative an/or anticoagulant potential and have been selected for further studies on structural features characterization as well as in vivo experiments are needed and are already in progress.
