SUMMARY:

Diabetes mellitus (DM) is a chronic metabolic disorder characterized by persistent hyperglycemia due to a deficiency in insulin production by the pancreas and/or in its action, causing long-term complications. Pharmacological therapy for DM has many limitations due to the high number of side effects and ineffective action against its complications, including nephropathy, the second main complication of DM and one of the most worrying for public health. Thus, alternative treatments based on medicinal plants are potentially useful for DM therapy. The species Spondias tuberosa Arruda, popularly known as the umbuzeiro, has been widely used in traditional medicine to treat digestive diseases, infections and diabetes, but it has been little explored scientifically. Considering its pharmacological potential, the aim of this study was to evaluate the anti-diabetic effect of Spondias tuberosa Arruda (ExSt) extract on diabetic nephropathy using biochemical, morphological, stereological and molecular analyses in diabetic rats treated with this species. 45 wistar rats were used and DM was induced by streptozotocin (40mg/kg i.p.). The groups were divided into: CT: non-diabetic animals; DM: animals with experimental DM; EX: non-diabetic animals treated with S. tuberosa; DMEX: animals with experimental DM treated with. S. tuberosa and DMIN: animals with experimental DM treated with insulin. ExSt (500 mg/kg, v.o.) was administered daily for 30 days after the onset of DM, as was insulin. Blood and kidney samples were collected for analysis. Among the results observed, ExSt showed an improvement in diabetic nephropathy, since parameters such as fibrosis, tubular necrosis and the amount of EROS were minimized, as well as improvements in other parameters such as haemorrhage, inflammation and corpuscular volume. Oxidative/antioxidant enzymes and immunohistochemistry of renal injury will be carried out to better understand the action of ExSt in experimental diabetic nephropathy, thus encouraging the search for new forms of treatment using natural compounds available in Brazil's rich and diverse flora.
