RESUMO:

Phytol is a diterpene alcohol that reduces cytokines, oxidative stress and inhibits the inflammatory response. In this study, we describe the preparation and characterization of non-cationic and cationic solid lipid nanoparticles (SLN) functionalized with polyethyleneimine and incorporated with phytol. The formulations were prepared with 1,3-distearyl-2-oleyl glycerol (TG1) natural lipid by solvent evaporation emulsification method and the physicochemical properties, biocompatibility and antifungal efficacy were evaluated for the different samples. SLN were monitored using dynamic light scattering, atomic force microscopy and drug encapsulation efficiency and the results showed that all SLNs have size <300 nm, PdI <0.2 and encapsulation efficiency> 65%. Our results demonstrate that phytol-free SLNs were not cytotoxic to HEK-293, while phytol-SLNs had a dose-dependent cytotoxic effect. The minimum inhibitory concentration assay was used to evaluate the in vitro anticandidal activity of SLN against seven reference strains and eight clinical isolates of Candida species. SLN were able to improve phytol activity and for reference strains MICs 50% were obtained with free phytol in solution (2500-625 µg/mL) and with 10-SLN-PHY (15.62-1.95 µg / mL), while in clinical isolates there was no activity. On the other hand, 5-SLN-PHY and 3-SLN-PHY samples produced MICs 100% at 0.24 and 0.40 µg/mL, respectively, for both types of strains. The results for C5-SLN-PHY were also equivalent to 0.24 and for clinical strains, this presented MICs 100% ranging from 7.81 to 0.24 µg/mL.
