SUMMARY:

Seed germination and seedling establishment are critical processes for plant communities, as high mortality rates take place during these first moments of the plant life cycle due to the exposition to biotic and abiotic stresses. Considering that plant survival depends on carbon metabolism in the transition from heterotrophy to autotrophy, this project aims to investigate the utilization of reserves in growth and the partitioning of non-structural carbohydrates in the different organs of Erythrina velutina Willd., an endemic pioneer species of the Caatinga, during seed germination and seedling establishment, verifying changes in the source-sink relations. In this way, seeds from ten mother trees will be scarified, disinfected, imbibed in distilled water, sown in towel paper rolls and maintained under controlled for 9 days. After that, seedlings will be transferred to distilled water in hydroponics and cultivated at a greenhouse for 12 days. The harvests will be carried out at distinct physiological stages, including imbibed seed, radicle protrusion, hypocotyl emergence, hook opening, cordiform leaf emergence, first trifoliolate leaf sprout, and second trifoliolate leaf sprout. Seedlings will be divided in cotyledons and other parts, according to their appearance during the different stages. Growth will be evaluated by dry weight accumulation in the different parts and the root/shoot ratio. The contents of soluble sugars, non-reducing sugars, and starch as well as the activities of amylases, neutral invertases, and acid invertases will be determined in each seedling part. In the course of the experiments, the harvests will be randomly performed to obtain five replicates for each physiological stage and determination. The experimental unit will correspond to a seedling and the results will be analyzed according to descriptive statistics using mean and standard deviation. Dry weight accumulation in the different seedling parts varied in the course of the establishment process. It was observed high dry weight investment in the shoots than in the roots, evidenced by the decrease of the root/shoot ratio during the experiment. In relation to the total soluble sugars and non-reducing sugars, all seedling parts demonstrated high content during their elongation or expansion.
