SUMMARY:

Diabetes Mellitus (DM) is a complex disease that requires continued medical assistance to reduce risk factors in addition to glycemic control. Among the main complications of DM are micro and macrovascular disorders related to the renal, cardiovascular and nervous systems. Hyperbaric oxygen therapy (OTH) involves intermittent administration of 100% O² under high pressure, which could resolve damage from hypoxemia and tissue hypoxia in nerve lesions. Objective: The ain this study is analyze the effects of OTH on sciatic nerve regeneration in diabetic rats. Methods: sixty male Wistar rats weighing about 220-300g will be divided into four groups: C (normoglycemic animals) C+L (normoglycemic animals with sciatic nerve injury), DM + L (diabetic animals with sciatic nerve injury) DM + L + OTH (diabetic animals with sciatic nerve injury and submitted to OTH). The animals of the diabetic groups will undergo induction of DM by Streptoozotocin (STZ) and the animals of the groups with injury will be subjected to induction of injury in the sciatic nerve. For the treatment protocol, the animals will be submitted to OTH with pressure inside Of the chamber of 3 absolute atmospheres (ATA) at a rate of 2 ATA / min and maintained at that pressure for 60 min. The chamber will be ventilated with 100% O 2 at a flow rate of 20 L / min in order to minimize CO2 accumulation. Morphological variables will be evaluated by means of histological, electrophysiological (CMAP peak amplitude, onset CMAP latency and conduction velocity (VCN)) and functional techniques using sciatic function index (SFI). Data will be analyzed using SPSS 22.0 software, with a significance level of 5%. The normality of the data will be verified by the Kolmogorov-Smirnov test and then the corresponding statistical tests will be applied. Outcomes: it is expected that the objectives will be achieved and that the results will have clinical and scientific relevance.
