SUMMARY:

The circadian timing system has evolved to allow organisms to synchronize their
physiology and behavior with the 24-hour rhythms in the environment. The ability of
light to reset or phase shift the circadian pacemaker is illustrated by the effects of light
on the circadian phase when the pacemaker is functioning freely in an environment
without timing signals. This system, in rodents, is composed of interconnected neural
structures, the suprachiasmatic nucleus (SCN) of the hypothalamus. There are several
major pathways that project to the SCN that allow it to synchronize with the 24-hour
environment, including: the retinohypothalamic tract (TRH), a direct projection of the
retina geniculohypothalamic tract (TGH), a direct projection from the intergeniculate
leaflet (FIG), and a direct serotonergic projection (5-HT) from the median raphe.
Research with psychedelics has grown and gained prominence in several areas of study.
Among the substances of greatest interest today, N,N-Dimethyltryptamine (DMT)
stands out, the main psychoactive present in Ayahuasca, a decoction of indigenous
origin from the peoples of the Amazon Basin, extracted from Banisteriospis caapi and
Psychotria viridis. These compounds are characterized by a general ability to induce
mood swings, variations in a person's perception, primarily visual, and an altered state
of consciousness, as well as intellectual or spiritual insight, similar to what is
experienced only through dreams or in moments of religious exaltation. This substance
is an agonist of the serotonergic system, promoting changes in brain functions. Anxiety
and mood disorders, as well as depression, are among the most disabling and
debilitating diseases around the world, negatively and substantially impacting global
societies, generating high costs for health systems. We aimed, with this project, to
evaluate the effects and possibly the impacts of DMT on the circadian timing system, as
well as to evaluate the activity/rest, and possible behavioral alterations, in animals,
formerly submitted to the administration of DMT, and consequently to evaluate through
from immunohistochemistry, possible alterations in biomarkers such as neuropeptide Y
(NPY), Brain-Derived Neurotrophic Factor (BDNF) and cFOS in the rodent brain, in
the regions of the Prefrontal Cortex (PFC), FIG and Pregeniculate Nucleus of the
Thalamus (PNT).
