Ethnopharmacological relevance
Hancornia speciosa (Apocynaceae), popularly known as “mangabeira,” has been used in folk medicine to treat inflammatory disorders, hypertension, dermatitis, diabetes, liver diseases and stomach disorders. Regarding the Hancornia speciosa fruit, the ethnobotany indicates its use especially for treating ulcers and tuberculosis. However, no study has been done so far to prove such activities.
Aim of the study
Thus, this study was designed to identify bioactive molecules in the aqueous extract of Hancornia speciosa (Mangabeira) fruit and also to investigate the ability of this extract and isolated compounds in inhibiting the inflammatory process.
Materials and methods
Aqueous extract of Hancornia speciosa fruit was prepared by decoction and the secondary metabolites were identified by high performance liquid chromatography (HPLC). The anti-inflammatory properties of the aqueous extract and isolated compounds of Hancornia speciosa were investigated using in vitro and in vivo models. In vivo tests comprised the xylene-induced ear edema, carrageenan-induced peritonitis and zymosan-induced air pouch (leukocytes count, cell migration kinetics and differential cell count), whereas in vitro tests consisted of determining the levels of cytokine IL-1 β, IL-6, IL-12 and TNF-α using ELISA.
Results
Rutin and chlorogenic acid were detected in the aqueous extract by HPLC. Furthermore, the aqueous extracts as well as rutin and chlorogenic acid significantly inhibited the xilol-induced ear edema and reduced cell migration in the animal models such as carrageenan-induced peritonitis and zymosan-induced air pouch. In addition, reduced levels of pro-inflammatory cytokines were observed.
Conclusion
This is the first study that demonstrated the anti-inflammatory effect of the Hancornia speciosa fruit against different inflammatory agents in animal models, suggesting that these extracts and/or their bioactive molecules, especially rutin and chlorogenic acid, have potential use in the inflammation therapy and therefo
