Passiflora edulis var. flavicarpa Degener aerial parts (leave and fruit) are frequently employed in folk medicine as ansiolytic and sedative. The pericarp of the fruit is now being investigated for medicine purposes. Scarce pharmacological data is avaiable and there are no reports about it toxicity. The aim of the present study was investigate the sub chronic toxicity in male rats and evaluate the reproductive toxicity in pregnant rats and exposed fetuses of an extract obtained by infusion of the pericarp in water (1:3 m/v;100o C) during 10 min. The extract composition was evaluated by tube reactions and thin lawyer chromatography (TLC). Adult male rats (n=8) were treated with 300mg/kg of the extract, by gavage, during 30 days and pregnant rats (n=7) were treated with the same dose from gestation day 0 to gestation day 20. Control rats (same number/gender) received tap water (1mL) by same conditions. Water and food intakes and body weight gain were recorded during treatment. At day 29 of treatment the sexual behavior of the males was analyzed and then half of males from each group received cyclophosphamide (50mg/kg, i.p.) for the (anti)genotoxic assessment of the extract by counting micronucleated polichromatic erythrocytes in bone marrow. Next day, after treatment with the extract, the males were anesthesized for blood (biochemical parameters: AST, ALT, urea and creatinine; hematological parameters: leukocyte counting), bone marrow and organs collection (histopathological study of liver, kidney, spleen, pancreas, heart and testis). At day 20 of gestation, the dams were anesthesized and utherine horns were exteriorized, after blood and organs collection. The reproductive performance was evaluated by recording parameters as number of fetuses/utherine horn, number of fetuses/litter, number of corpora lutea/ovary, number of alive and dead fetuses. The fetal analysis was conducted by visceral and skeletal examination of four fetuses from each litter per technique employed. The phytochemical analysis revealed the presence of flavonoids, unspecific alkaloids, phenolic and triterpenic compounds in the extract. The statistical analysis revealed absence of significant differences between experimental and control groups for all the parameters evaluated for males and females. The extract also showed no (anti)genotoxic effect or capacity to alter clastogenicity induced by cyclophosphamide. No visceral or skeletal malformations were detected in exposed fetuses. This study suggest that the aqueous extract obtained from pericarp of P. edulis var. flavicarpa Degener was not able to promote toxic effects in adult male and female pregnant rats treated orally, and fetuses exposed during gestation, to 300 mg/kg/day at the treatment periods used.
