SUMMARY:

Clinical evidence indicates that deficiencies in the nucleotide excision repair (NER) and base excision repair (BER) pathways are associated with neurodegenerative pathologies in humans, suggesting that these pathways play a critical role in chronic neurodegenerative diseases. Failure in the NER pathway results in various DNA damages, such as continuous activation of poly(ADP-ribose) polymerase 1 (PARP1) and decreased intracellular NAD+. However, the relationship between mitochondrial dysfunction and BER pathway deficiency, as well as the potential interaction with the NER pathway, is not yet fully deficiencies in the ubiquitin-proteasome system (UPS) and mitochondrial metabolism.
Thus, the study evaluated how deficiencies in EXO-3 and XPA-1 affect mitochondrial integrity and the development of neurodegenerative phenotypes in transgenic C. elegans models. Knockdown animals for exo-3 and/or xpa-1 from strains expressing GFP in intestinal and muscle mitochondria were analyzed, and the effect of BER and NER pathway deficiencies on apoptosis was also observed.
The results support previous findings regarding increased mitochondrial content in exo-3;xpa-1 knockdown animals. The focus now shifts to the development of mutant animals with exo-3 and/or xpa-1 deletions for a better characterization of mitochondrial function and neuronal integrity. It is expected that this study will contribute to a better understanding of neurodegenerative diseases and the role of EXO-3, a central endonuclease in BER repair, in the emergence of neurodegenerative phenotypes and its association with mitochondrial dysfunction.
