SUMMARY:

The molecular events involved in the heterogeneous behavior of odontogenic lesions are still not well understood. It is known that pathways that involve the mechanisms of the cell cycle and apoptosis are affected during the process of tumorigenesis. INGs (inhibitor of growth gene) proteins play the role of tumor suppressors regulating important cellular functions such as chromatin remodeling, cell proliferation, DNA replication and repair, cell migration and angiogenesis. This work proposes to analyze, through immunohistochemistry, the correlation between the expression of proteins of the ING family (Ing1, Ing2, Ing3, Ing4 and Ing5) and the expression of proteins regulating the cell cycle (p53) and apoptosis (Blc- 2 and Bax) in benign odontogenic lesions of epithelial origin. The sample will be intentional and non-probabilistic, for convenience, consisting of 20 cases of ameloblastomas, 20 cases of adenomatoid odontogenic tumors, 20 cases of odontogenic keratocysts and 10 samples of dental follicles. Initially, the morphological analysis will be performed to describe the histopathological aspects of the lesions that will compose the sample of the present study. Then, specimens will be subjected to marking by immunohistochemical method, using antibodies anti-Ing1, anti-Ing2, anti-Ing3, anti-Ing4, anti-Ing5, anti-p53, anti-Bax and anti- Bcl-2. For the evaluation of immunostaining, it will be a quantitative analysis taking into account the cell location (cytoplasmic, nuclear or nucleocytoplasmic). Five representative fields of the high magnification lesions will be photographed, and then, with the aid of the ImageJ® program, the positive and negative cells for the studied antibodies will be counted, evaluating the nuclear and cytoplasmic compartments separately, and thus, the percentage will be established. of cells exhibiting cytoplasmic and nuclear immunopositivity for each case. The results obtained will be typed in an electronic spreadsheet and analyzed with the aid of the Statistical Package for the Social Sciences software. For all statistical tests used in the present study, the level of significance will be set at 5% (p <0.05) and analyzes will be carried out to verify the hypotheses.
