ISSR MOLECULAR MARKERS FOR GENETIC DIVERSITY STUDIES OF Handroanthus impetiginosus

Handroanthus impetiginosus is a tree species with high ecological and economic potential. In the Brazilian market, wood is heavily exploited in illegal trade. Therefore, studies on genetic diversity with the standardization of species conservation are necessary. Thus, this study aimed to select Inter Simple Sequence Repeat (ISSR) primers for studies of genetic diversity applied to forest populations of H. impetiginosus. For that, 30 ISSR molecular markers were tested in 30, evaluating the total number of loci, polymorphism rate and content value of the polymorphic information, as well as the marker index and the resolving power. The primers were selected for presenting the best amplification pattern, which provided 62 loci and 61 polymorphic loci. The value of the information content of the polymorphic primers varied from 0.34 to 0.49, while the value of the marker index (MI) obtained an average of 3.20, resolving power (RP) of 2.40, diversity of Nei (He) of 0.35 and the Shannon (I) index of 0.52. The data demonstrate are ISSR markers 807, 809, 818, 824, 857, 860, 873 and 881 are efficient for the quantification of the genetic diversity of H. impetiginosus. These results can support strategies aimed at species conservation and matrix selection.
